Study of Triterpene Saponin Compounds from Centella asitica as Renin Inhibitor with Pharmacophore Modeling, Molecular Docking and In-vitro Evaluation

Hypertension is a silent killer that causes kidney, heart, and stroke damage if not handled properly. In Indonesia, the prevalence of population with high blood pressure 34.11% women 36.85% higher than men 31.34%, this shows fairly value so special attention needed on hypertension therapy. It known currently there are 6 types pharmacological therapy for one newest renin inhibitor class (Aliskiren). Indonesia has diverse natural wealth in form flora fauna, more 30,000 medicinal plants 9500 potential herbal medicines have been utilized optimally, largest exporter world. Centella asiatica containing triterpenoid saponins activity, namely content Asiaticoside Madecasoside. The research method was carried out silico using molecular simulation vitro fluorometry (328/552 nm) to test activity asiaticoside madecasoside compounds as well mixture plants. This supported by docking outcome. results show madecososide gibbs energy close positive control aleskiren (-8.356 kcal/mol) (-9.44 kcal/mol). experiment showed saponin compound (madecassoside) contained an IC 0.71, at concentration 5 μg/μl, absorbance 1.35 A first minute. strongest inhibition Madecasoside μg/μl average fluorescent adsorption percent 135% best ug/ul minute values 1.19 A. Finally, result corresponded experiment. antihypertensive, especially secondary metabolites triterpene pure compared aliskiren inhibitor. So antihypertensive.
